Abstract
Drug formulation success is highly dependent on factors such as solubility, stability, and compatibility with excipients. A significant challenge in drug development is the poor solubility of nearly 40% of novel chemical entities, which limits their bioavailability and therapeutic efficacy. Traditional formulation strategies, including micronization, salt formation, and the use of surfactants, have been employed to address these issues, but they often lack universal applicability. Nanotechnology has emerged as a promising solution, particularly through the development of nanosuspensions, which are submicron colloidal dispersions of drug particles stabilized by surfactants. Nanosuspensions enhance the solubility and bioavailability of poorly water-soluble drugs by reducing particle size to the nanoscale, leading to faster dissolution rates and improved absorption. This review discusses various methods for preparing nanosuspensions, such as bottom-up and top-down technologies, including precipitation, high-pressure homogenization, and supercritical fluid methods. Additionally, it highlights the applications of nanosuspensions in oral, parenteral, pulmonary, ocular, and targeted drug delivery systems. Marketed nanosuspension-based drugs and their routes of administration are also reviewed, demonstrating the versatility and effectiveness of this technology in improving drug delivery. Overall, nanosuspensions offer a robust approach to overcoming the challenges associated with poorly soluble drugs, paving the way for more effective therapeutic interventions.
