Abstract
Quercetin is an occurring flavonoid that has attracted a lot of attention in pharmaceutical research but it has a big problem. It does not dissolve well in water, which makes it hard to formulate. Nanostructured lipid carriers, which are made from a mix of liquid lipids and kept stable by surface-active agents offer a good way to include compounds that do not mix well with water and release them in a controlled way. This extended paper brings together the work done on formulating Quercetin in the M.Pharm dissertation and a discussion from the literature about delivering Quercetin, the structure of NLCs how to give them through the nose the variables in the formulation how to test them and the challenges of taking them to life. A method based on screening was used to create and test ten formulations. F10, which includes Quercetin, Compritol 888 ATO MCT oil, Tween 80 and Poloxamer 188 had the physical and chemical properties. The size of the particles was 142 ± 2.6 nm the PDI was 0.21 the zeta potential was −30.2 mV the encapsulation efficiency was 91.5 ± 0.8%. The drug loading was 8.5 ± 0.2%. After 8 hours the total release was 84.2 ± 3.1% and the first-order model had the R² value for F10. Testing for three months showed some changes in the size of the particles the PDI, the zeta potential and the amount of drug. There was no separation of layers or any precipitation. Research shows that more study is needed on lipid nanocarriers for delivering medicine through the nose. The current work did not show that the medicine could pass through the nose, how it moves in the body, where it goes if it targets the brain or if it works as a treatment. So F10 should be seen as a candidate, for more research before it can be considered as a way to target the brain or as a treatment that works.
