The aerial portion of 1ymus schimperi is frequently used in Ethiopian traditional medicine to treat conditions such as fungal skin infections, gonorrhoea, cough, liver and renal disorders, hypertension, and stomachaches. The purpose of this study was to assess the acute, subacute toxicity of 1ymus schimperi essential oil in Wistar albino rats, as there has been insufficient research on the toxic effects of this oil. Approach. Following hydro distillation, the essential oil of the aerial portion of T. schimperi was subjected to toxicity tests and GC-MS analysis. Rats were randomly assigned to seven groups (n = 5) for the Dermal toxicity investigation. The experimental groups were given single doses of 300, 600, 900, 1200, 1500, and 2,000 mg/kg of the oil, whereas the control group was given just distilled water with 2% Tween 80. Blood samples were taken for hematological and clinical chemistry analysis after the trial. The liver and kidneys' gross pathology and histopathology were also assessed. Additionally, vNN and ADMET servers were used to predict the toxicity parameters. outcomes. The T. schimperi essential oil's LD50 in this investigation was determined to be 1284.2 mg/kg. The oil is categorized by the World Health Organization as moderately dangerous when taken orally. Furthermore, no significant differences were observed between the control and essential oil-treated groups, observed in the gross histopathology of the liver and the kidneys. While some of the chemicals displayed hepatotoxicity and mitochondrial membrane potential toxicity, the majority of the oil's constituents were shown to be harmless in the in-silico toxicity research. Therefore, before utilizing compositions containing the essential oil of T. schimperi, it is imperative to do a chronic toxicity analysis of the essential oil and its constituents, which showed toxicity in the in-silico study.
