Abstract
Efficient extraction of high-quality genomic DNA is pivotal for genetic research, particularly in plant species with challenging sample types such as dry, fibrous tissues. This study employed a modified Qiagen DNeasy Plant Mini Kit protocol to enhance DNA yield and purity from 37 grass species collected across diverse regions of India. Grass species play a critical role in ecosystems and agriculture, making their genetic analysis essential for understanding biodiversity, adaptation mechanisms, and evolutionary dynamics. The modifications introduced in this study addressed issues such as low DNA yield and contamination, improving the extraction process for dried plant samples. DNA purity and concentration were assessed using NanoDrop spectrophotometry and agarose gel electrophoresis, revealing significant improvements over conventional methods. Results highlight the protocol's effectiveness in obtaining high-quality DNA for downstream applications, including phylogenetic analysis and conservation research. This work contributes to advancing genetic studies on grass species, crucial for ecological balance and agricultural development.
