Abstract

Herbal plants and products represent important source of natural antioxidant and offer an alternative to synthetic protectors. The major use of herbal medicines is for health promotion and therapy for chronic, as opposed to life-threatening, conditions. However, usage of traditional remedies increases when conventional medicine is ineffective in the treatment of disease, such as in advanced cancer and in the face of new infectious diseases. Furthermore, traditional medicines are widely perceived as natural and safe, that is, not toxic. Picrorhiza kurroa, a herbal medicinal plant has been screen for their radioprotective activity. For the purpose of study male Swiss albino mice were divided into seven groups. Group I serve as normal. Group II to IV were exposed to gamma radiation (2.5 Gy and 5.0 Gy) and also provided mercuric chloride (0.5 ppm) both separately and combined. The herbal radioprotector Picrorhiza kurroa (kutki) was given to experimental groups (V to VII) after irradiation and mercury treatment. The drug was given seven days prior to treatment with radiation. Animals were autopsied at post treatment interval of 1,2,4,7,14 and 28 days. After sacrificing the animal’s pieces of kidney was kept at -20oc for the estimation of various biochemical parameters such as Total proteins, Glycogen, Cholesterol, Acid Phosphatase activity, Alkaline phosphatase activity, DNA and, RNA.It was found that Total proteins, Glycogen, Acid phosphatase, Alkaline phosphatase and RNA content of renal cells was increased while Cholesterol and DNA was decreased. Renal biochemistry depicted degenerative changes and changes were more pronounced in combined treatment groups and show synergistic changes. Picrorhiza treated groups demonstrated early and fast onset of recovery against deleterious effects of radiation and mercury in comparison to non-kutki treated groups. Therefore, present study shows free radical scavenging and antioxidant properties of Picrorhiza which was due to various glycosides reported in kutki.
