Abstract

ABS TRACT Escherichia coli is a Gram-negative, facultative anaerobic and rod-shaped bacterium of the genus Escherichia, found in the lower gastrointestinal tract of warm-blooded animals. E. coli is a fast-growing bacterium which can easily mutate due to horizontal gene transfer which ensures its evolutionary distribution and strain diversity within the environment. The aim of this research work was to determine the prevalence of Escherichia coli isolated from diverse samples within Abakaliki Metropolis of Ebonyi State, Nigeria. A total of one hundred and eighty-one (181) samples were collected from abattoir effluent, kunu and urine as follows: twenty-five (25) abattoir effluent samples, thirty-six (36) kunu samples and one hundred and twenty (120) urine samples. Escherichia coli strains were isolated using standard microbiological techniques on MacConkey agar plates, while morphological and biochemical characteristics were employed in identifying the E. coli strains. Antimicrobial susceptibility test was carried out using Kirby and Boar disc diffusion and 0.5% McFarland turbidity standard as described in the clinical laboratory standard institute. Molecular characterization involved polymerase chain reaction and gene sequencing. The result showed that all the twenty-five abattoir samples collected, E. coli was recorded in all, out of the thirty-six kunu samples, E. coli was recorded in thirty-three samples, of the one hundred and twenty urine samples collected, E. coli was recorded in twenty-two samples. E. coli strains isolated from the three diverse samples showed close susceptibility pattern to the tested antibiotics with the highest susceptibility rate to Ciprofloxacin, Gentamycin, and Trimethroprin/Sulphamethoxazole, and total resistance to Tetracycline and Imipenem. The result of the Molecular analysis recorded diverse strains of E. coli from the diverse samples.
