Abstract
ABSTRACT The study aimed to prepare and evaluate a wintergreen oil cream for anti-inflammatory activity using the egg albumin denaturation method. From ancient times Gaultheria procumbens L. essential oil was known for its medicinal properties. The primary component, methyl salicylate, was identified through chemical tests and thin-layer chromatography (TLC). The absorption maxima for the oil was determined using UV-visible spectrophotometry. The anti-inflammatory activity of the oil was assessed using the egg albumin denaturation assay, comparing the inhibition percentage to that of standard diclofenac sodium. The wintergreen oil exhibited distinct organoleptic properties, with a specific gravity of 1.175, viscosity of 0.2053 cP, ester value of 67.0466%, and a boiling point of 224°C. TLC confirmed the presence of methyl salicylate with an Rf value of 0.73. The absorption maximum for the oil was found at 236 nm. In the egg albumin denaturation assay, the oil demonstrated significant anti-inflammatory activity, with a maximum inhibition percentage of 90.40% at 500 μg/ml, performing both diclofenac sodium and methyl salicylate standards. A cream formulation containing 500 μg wintergreen oil was prepared and evaluated for organoleptic characteristics, homogeneity, colour, pH, and Spreadability. The cream was homogenous, light pinkish in colour, with a pH of 6.2 and Spreadability of 5.61 g.cm/s. The formulated wintergreen oil cream exhibited significant anti-inflammatory properties, suggesting its potential use as a topical anti-inflammatory agent. Further studies are warranted to explore its clinical efficacy and safety.
