Abstract
Donepezil HCl (DP) is an acetyl cholinesterase inhibitor used for the treatment of behavioural and cognitive effect of Alzheimer’s disease and other type of dementia. The DP-orodispersible tablets were made using direct compression method and mixture of superdisintegrant crospovidone (CP) and croscarmellose sodium (CCS) chosen through central composite design (CCD). The 23 factorial designs with three responses viz., dispersion time, disintegration time and t50. The precompression results show desirable compression properties and FTIR studies no interaction between the DP and added excipients. The eleven trials of tablets were made and evaluated their response data, further numerical optimization was done through point prediction method using Design expert software Trial V13 to generate optimized formula. The optimized formula was made into tablets, evaluated and validated through statistical methods. The overall results conclude that design of experiments studies can be conveniently applied for optimization of DP loaded orodispersible tablets.
