Abstract
Obesity induces cardiovascular diseases through pressure or volume overload, mitochondrial dysfunction, cardiac fibrosis and apoptosis. The aim of this work is to study the light and electron microscopic changes in the heart caused by obesity in Wistar albino rats. Sixty male albino rats (120 ± 5 gram and 5 weeks old) were divided into: Control group: fed normal laboratory diet 4 weeks. Treated group: treated by intra-peritoneal injection of Triton (250 mg/kg) to induce obesity and fed normal laboratory diet 4 weeks. Recovery group: as treated group and left for another 4 weeks. At the end of the experiment, body weight and serum lipid profile were measured. Myocardial samples were taken for LM and EM study. LM slides were stained, photographed and analyzed. Ultra-thin sections were examined by transmition electron microscope. Means and standard error were calculated for body weight, serum lipid profile, myocardial cell perimeter and total area of interstitial collagen. Statistical analysis was done by ANOVA to compare between groups. Results: body weight, serum level of lipid profile and atherosclerotic indices showed significant increase in treated group then significant improvement in recovery group. Light microscopic study of treated group revealed marked increase in cardiac myocytes with disorganization of myofibrils by vacuoles. Infiltration of myocardium by adipocytes and collagen fibers are also seen. In recovery group, myocardial cells showed mild improvement in size however myofibrils still disorganized by vacuoles. Some fat cells are seen with mild decrease in collagen fibers. Statistical analysis of treated group revealed significant increase in both cardiac cell perimeter and total area of interstitial collagen fibers. However, recovery group showed significant improvement in cardiac cell perimeter and non-significant improvement in total area of interstitial collagen fibers. Electron microscopic study of treated group revealed marked increase in interstitial collagen fiber and adipocyte infiltration. Disorganization of myofilaments by large lipid droplets is present in multiple areas. Mitochondrial rupture with decrease in their number and swelling in their cristae are also seen. Recovery group revealed slight improvement in both interstitial collagen and adipocyte infiltration. myofilaments showed moderate improvement and intracellular fat droplet disappeared. Mitochondria started to increase in number, however some mitochondrial rupture and swollen cisternae still present. Conclusion: rats treated with triton exhibited obesity features characterized by increased body weight, elevated serum lipids and histological cardiac abnormalities. So, healthy diet program should be recommended to avoid obesity and obesity-related cardiac complication.
